Claims Scope:

Patent issued December 21, 2023. 

Sequence agnostic composition (non-integrating viral vector and pharmaceutical composition) and method (expressing a gene and selecting an optimized non-integrating viral delivery system) claims comprising: 

• A viral carrier with defective integrase gene 
• Human HPV viral episomal origin of replication with a 5’ truncated HPV 16 LCR 
• Sequence encoding an initiator protein specific for viral episomal origin of replication 
• A gene, gene product, shRNA, siRNA, miRNA, or other RNA 

Wherein the 5’ truncated HPV 16 LCR is selected from: 

• Nucleic acid encoding an E1 and two E2 binding sites 
• Nucleic acid encoding an E1 and three E2 binding sites 
• Nucleic acid encoding two AP1 and an E1 binding sites 
• Nucleic acid encoding three AP1 binding sites
