Claims Scope:

Sequence agnostic composition (non-integrating viral vector/pharmaceutical composition) and method (expressing a gene by contacting cells or administering to a subject/ non-integrating viral vector with HPV 16 5’ truncated LCR specific for each of Quadrants 1-4) claims comprising: 

• A viral carrier with defective integrase gene 
• Human HPV viral episomal origin of replication with a 5’ truncated HPV 16 LCR 
• Sequence encoding an initiator protein specific for viral episomal origin of replication selected from E1, E1-C, E2, E2-11, E1-T2A-E2 and EBNA-1 
• A gene, gene product, shRNA, siRNA, miRNA, or other RNA 

Wherein the 5’ truncated HPV 16 LCR is selected from: 

• Nucleic acid encoding an E1 and two E2 binding sites 
• Nucleic acid encoding an E1 and three E2 binding sites 
• Nucleic acid encoding two AP1 and an E1 binding sites 
• Nucleic acid encoding three AP1 binding sites
